What is an Aceclofenac Tablet?   

Aceclofenac is an anti-inflammatory drug. It is a nonsteroidal or NSAID, analog of diclofenac. It is used for relieving any kind of pain, fever or inflammation in numerous medical cases; as it potently inhibits the cyclo-oxygenase enzyme (COX) that is involved in the synthesis of prostaglandins. 

It is reported to have a higher anti-inflammatory action or at least comparable effects than conventional NSAIDs in double-blind studies, and belongs to BCS Class II as it possesses poor aqueous solubility. It is practically insoluble in water with good permeability.

Aceclofenac (C16H13Cl2NO4), being chemically [(2-{2, 6-dichlorophenyl) amino} phenylacetooxyacetic acid]; is a crystalline powder having a molecular weight of 354.19. 


How do Aceclofenac Tablets work? 

In the simplest of terms, Aceclofenac tablets work by inhibiting the action of cyclooxygenase (COX) that is involved in the production of prostaglandins (PG) which is accountable for pain, swelling, inflammation and fever. 

Through COX-2 inhibition, aceclofenac checks the production of multiple inflammatory mediators like prostaglandin E2 (PGE2), IL-1β, and TNF from the arachidonic acid (AA) pathway. Inhibition of IL-6 is considered to be mediated by diclofenac converted from aceclofenac. Further, the suppressed action of inflammatory cytokines decreases the production of reactive oxygen species. 

Aceclofenac is found to have reduced the production of nitrous oxide in human articular chondrocytes. In addition, aceclofenac interferes with neutrophil adhesion to endothelium by decreasing the expression of L-selectin (CD62L), which is a cell adhesion molecule expressed on lymphocytes. 

This drug is proposed to stimulate the synthesis of glycosaminoglycan in human osteoarthritic cartilage which may be mediated via an inhibitory action on the IL-1 production and reaction. 

The chrondroprotective effects are generated by 4'-hydroxyaceclofenac which suppresses IL-1 influenced production of promatrix metalloproteinase-1 and metalloproteinase-3, and interferes with the release of proteoglycan from chrondrocytes. 


When to avoid Aceclofenac? 

People with porphyria or breast-feeding mothers, and children should be kept away from using Aceclofenac. It should also be avoided by any pregnant woman because of the high risk of having a premature closure of ductus arteriosus leading to fetal hydrops in the neonate.
