What is Menthol?
Menthol is an organic compound, or rather a monoterpenoid, that can be made synthetically or be obtained from the oils of corn mint, peppermint, or other mints. It is a waxy, clear or white crystalline substance, which melts if kept at slightly above the room temperature.
The particular form of menthol that one may find in nature is (−)-menthol, which is expressed in the (1R,2S,5R) configuration. Menthol has local anesthetic and counterirritant qualities, and is used to treat various irritations apart from mild to moderate muscle and joint pain. Menthol also acts as a weak κ-opioid receptor agonist.
How does Menthol work?
Menthol primarily activates the cold-sensitive TRPM8 receptors in the epidermal layer or skin. After topical application, it causes a cooling sensation due to stimulation of 'cold' receptors via the inhibition of Ca++ currents of neuronal membranes. It may also yield some analgesic properties via kappa-opioid receptor agonism.
