What is Tolnaftate?   

Tolnaftate is a synthetic thiocarbamate used as an antifungal agent used to treat various skin infections caused by fungi, and can also be taken in combination with some other anti-fungal medicines. 

Tolnaftate is a thiocarbamate derivative with either fungicidal or fungistatic property. 

This pharmaceutical drug has been found to be slightly less effective than azoles when used to treat tinea pedis or athlete's foot. However, it has been found useful while dealing with ringworm, especially when passed from pets to humans. 

 

How does Tolnaftate work? 

Tolnaftate is a topical fungicide. Although its exact mechanism of action is yet unknown, it is believed to inhibit squalene epoxidase, an important enzyme in the biosynthetic pathway of ergosterol which is a key component of the fungal cell membrane, in a similar way to terbinafine. 

Chemically speaking, it is a selective, reversible and non-competitive inhibitor of membrane-bound squalene-2,3- epoxidase, an enzyme involved in the biosynthesis of ergosterol. This inhibition culminates into the accumulation of squalene and a deficiency in ergosterol, an essential component of fungal cell walls, which eventually increases the membrane permeability, disrupting cellular organization and causing cell death. Additionally, this agent may also distort the hyphae and stunts mycelial growth in susceptible fungi. 

In simple terms, it is supposed to prevent ergosterol biosynthesis by inhibiting squalene epoxidase. It has also been reported to distort the hyphae and to stunt mycelial growth in susceptible organisms.
